      Introduction
      • AutoPure Total Protein is a reagent kit for direct quantitative determination of total protein in human serum and plasma on automated clinical chemistry analyzers.
      • AutoPure Total Protein is ready to use, single liquid reagent system.
      • With AutoPure Total Protein the assay is linear upto 18 g/dl (180 g/l).
      Principle

      Divalent copper ion reacts in alkaline solution with protein peptide bonds to form purple coloured biuret complex.

      Protein + Cu2+ - Cu - protein complex. 

      The intensity of purple colour formed is directly proportional to the concentration of the total protein in the specimen and is measured photometrically.

      Reagent Storage, Stability & Handling

      AutoPure Total Protein is ready to use, single liquid reagent system.

      Shelf life

      Stable till the expiry date indicated on the label, when stored at 15-25 C.

      On  Board Reagent Stability.

      45 days at 2- 8C after opening. Protect the reagent from light and contamination.

      Do not freeze the reagent.

      Specimen Collection & Preservation
      Collect sample using standard sampling tube. Serum, heparinized plasma or EDTA plasma can be used. Using plasma, results could be falsely elevated upto 0.4 g/dI due to fibrinogen. Avoid haemolysis since haemoglobin reacts as protein in the total protein assay.

      Total protein in serum / plasma is stable for 3 days at 2- 8C and 6 months at  -20C. Centrifuge samples containing precipitate before performing assay.

      Calculations

      Fully automated system automatically calculates the total protein concentration of each sample.

      Results in g/I = Results in g/dI x 10

      For calibration, it is recommended to use C.f. a.s or any other suitable calibrator material.

       Quality Control

      Each batch of AutoPure Total Protein is assayed with atleast six quality control sera   Precinorm, Precipath, Biorad I , Biorad 11, Accutrol Normal and Accutrol Abnormal prior to release

      To ensure adequate quality control, it is recommended that the laboratory should use a normal and abnormal commercial reference control serum. It should be realized that the use of quality control material checks both the reagent and instrument functions together.

      If the control values fall outside the specified limits, each of the below criteria should be cross - checked and corrected:

       

      • Proper instrument function  wavelength setting, light source and temperature control.
      • Cleanliness of probes and cuvettes.
      • Bacterial contamination of wash water used by the instrument.
      • Expiry date of the reagent kit.

       

      Expected Values
      Serum/ Plasma
      Adults : 6.3 - 8.4 g/dI (63  - 84 g/1)
      Note:

      Expected range varies from population to population. It is therefore recommended that each laboratory should establish its own normal range. For diagnostic purposes, the total protein results should always be assessed in conjunction with the patient's medical history, clinical examinations and other findings.

      Performance Characteristics Linearity

      With AutoPure Total Protein, the assay is linear up to 18 g/dI (180 g/l). Determine samples with higher concentrations via the rerun function. On instruments without rerun function, manually dilute samples with higher concentrations using 0.9% NaCI or distilled/deionized water (e.g. 1 + 3). Multiply the result by the appropriate dilution factor (e.g. 4).

      Interference

      There is no significant interference in samples containing upto 40 mg/di of bilirubin and 500 mg/di of haemoglobin. Haemoglobin interferes in the results as it reacts as a protein in total protein assay.

       Co - Relation Studies

      A comparison of the Total Protein determination using AutoPure Total Protein reagent and AutoZyme Total Protein gave the following co relation (g/dI):

      Linear Regression

      y = -0.201 + 1.030x

      r = 0.996

      Sy.x = 0.10

      No. of samples measured : 85

      The sample concentrations measured were between 4 g/dI and 9 g/dl.
